Mechanism
Retatrutide is more potent than native GIP at the human GIP receptor, and less potent than the native hormones at the GLP-1 and glucagon receptors. In rodent models, the glucagon-receptor activity raises energy expenditure, adding to the appetite effects driven by the incretin receptors.
Key findings in the literature
01In obese mice, it reduced body weight more than tirzepatide at matched exposure, mainly through higher energy expenditure [1].
02A Phase 2 trial reported mean weight reductions of up to 24.2% at 48 weeks at the highest dose, compared with 2.1% for placebo [2].
